Interesują nas przede wszystkim badania oddziaływań białek oraz białek i substancji małocząsteczkowych w obrębie tematyki nowotowrzenia i patogenezy bakterii. Obecnie realizujemy projekty własne obejmujące charakterystykę strukturalną kinaz o istotnej roli w procesach nowotworzenia, szlaku białka p53, bakteryjnych proteinaz o nieznanym mechanizmie katalizy oraz proteinaz gronkowcowych. Równolegle do badań krystalograficznych rozwijamy także techniki selekcji aptamerów (szczególnie jednoniciowego DNA) przeciwko celom molekularnym istotnym w procesach nowotworzenia.
Poza badaniami własnymi prowadzimy także bogatą współpracę naukową oraz współpracę z przemysłem w zakresie innych tematów gdzie w rozwiązaniu problemów badawczych może być pomocna krystalografia rentgenowska białek. Zapraszamy do współpracy w tym zakresie.
We study different translation control mechanisms, which regulate the production of specific sets of proteins by chemical modifications of tRNA molecules. Every protein in the cell is produced by the ribosome, which uses transfer RNA (tRNA) molecules to translate the sequence information coded in mRNAs into correctly assembled poly-peptide chains. The decoding/translation of genetic information is based on the recognition of a respective codon by its corresponding tRNA anticodon triplet.
The lab is focusing on understanding the molecular mechanisms that lead to the specific base modifications in anticodons of tRNAs. These modifications have a strong influence on the efficiency and accuracy of the codon-anticodon pairing and therefore regulate the translational rates and folding dynamics of protein synthesis. Recent findings have shown that alterations of these modification pathways play important roles in the onset of certain neurodegenerative diseases and cancer.
HNF1A-MODY – Disease modeling in-a-dish
Maturity onset diabetes of the young (MODY) is caused by a mutation in a single gene and leads to diabetes under the age of 25. The mutations are inheritable, and any child have 50% chance of getting the same mutation as a parent. The mutations in HNF1A gene (leading to HNF1A-MODY) causes about 70% of all MODY cases. Diabetes is induced by lowering the amount of insulin. However, the clinical expression in HNF1A-MODY patients is quite variable even within the same family. Some can develop hyperglycemia, whereas others can be normoglycemic at the same age. It was shown that patients with HNF1A-MODY often develop diabetic microvascular complications, related to the endothelial dysfunction, however it is not clear whether these complications are result of the hyperglycemia or due to the genetic mutation in HNF1A gene.
Therefore, an improved understanding of the mechanisms and causes of endothelial dysfunction in HNF1A-MODY could provide new approaches for patient management. In the current project we are using two of the most impactful techniques for disease modeling –induced pluripotent stem cells (iPSCs) and CRISPR/Cas9 gene editing.
Our work is based on the usage of so-called isogenic lines that differ exclusively at the disease-causing mutation. Thus the individual variances could be diminished and subtle disease-relevant differences in monogenic diseases could be revealed.
Using isogenic iPSCs lines we found that cells with heterozygous mutation in HNF1A gene have increased vascular permeability after stimulation with pro-inflammatory cytokine (Kachamakova-Trojanowska et al., 2019). And this effect was even more pronounced in the cells with biallelic mutation. Currently, we would like to dissect the molecular background of the observed increase of the vascular permeability and to reveal its possible relation to HNF1A mutation.

Głównym obszarem badań naszej grupy jest fotobiologia, w szczególności wpływ światła krótkofalowego na funkcjonowanie roślin. Badania skupione są wokół fototropin, fotoreceptorów światła UV/niebieskiego. Drugi obszar badań naszej grupy stanowią zagadnienia związane z interakcją światła z liśćmi roślin. Specjalizujemy się w projektowaniu urządzeń do pomiaru właściwości liści, takich jak transmitancja i reflektancja. Trzecim z realizowanych tematów jest rozwój oprogramowania do analizy pomiarów AFM.
Nasze badania koncentrują się na tym, co wpływa na rozwój zarodka ssaków w perspektywie długoterminowej (Developmental Origins of Health and Disease, DOHaD). W kręgu naszych zainteresowań znajduje się wpływ technik wspomaganego rozrodu oraz starzenia się rodziców na stan zdrowia potomstwa w ciągu całego życia, a także transgeneracyjne dziedziczenie chorób.
