Project title: Structural Characterization of Targeted Protein Degradation Systems

Targeted Protein Degradation (TPD) describes the highly specific degradation of proteins that are responsible for promoting human pathologies. TPD combats the therapeutic limits of small molecules with overcoming drug resistance and uncoupling their pharmacokinetics from pharmacodynamics. TPD enables us to specifically target proteins that do not harbor any enzymatic function or active sites, which are typically not available for targeting by conventional small compound inhibitors. The common denominator among the currently available targeted protein degrading systems is the proximity-dependent chemical induction of E3 ubiquitin ligases, which mark their target proteins for destruction by the ubiquitin-proteasome pathway. Our project aims at using state-of-the-art structural biology techniques to enable informed design and selection of novel degraders, based on the geometry of ternary complexes formed between the E3 ligase, the target protein and the respective effector.
