Proteomics in neurobiology

The main expertise of the Proteomics and Mass Spectrometry Laboratory of MCB Jagiellonian University concerns large-scale, high throughput proteomic analysis in the field of neurobiology, particularly in the elucidation of mechanism of antidepressant and antipsychotic drug action. We use both gel-based two dimensional in gel electrophoresis (2D-DIGE) and mass spectrometry-based methods with tags like Isobaric Tags for Relative and Absolute Quantification (iTRAQ) as well as label-free technique. The objects of our investigations are different rat or mouse brain structures, model neuronal cell lines and primary glial cells.

Apart from the studies of the impact of stress and/or antidepressants on the protein profile of brain of laboratory animals and cells, one of the main topic we are working on is a characterisation of immunological background of Major Depression Disorder. In this project we look for differences in proteome of special subpopulation of T lymphocytes isolated from depressed patients and healthy volunteers.

The important stage of this project was elaboration of reproducible protocols of isolation of very low abundant fractions of T cells. In next step it was developed a procedure of protein extraction, purification and digestion of as tiny amount of material as 1 ug of total protein mixture. These studies are conducted in different group of patients like: first episode patients, patients in relapse, patients with chronically occurring symptoms. The project is carried out in collaboration with clinical psychiatrist.

Additionally, we collaborate with many groups on proteomic characterization of some biological aspects of different cell types, stem cells, cellular microvesicles, cancer cells and others.

Future plans

In future, we are going to focus on studies concerning subproteome of nuclear proteins in facet of mechanism of action of individual antipsychotic drug as well as in adjunctive therapy.
